Tim,

This seems to be the same problem I described a few days ago. It does appear
the file is locked by DBISAM even though the are no sessions active. In my
case the lock gets cleared when DBISAM is stopped and restarted.

Yes, but in his case the issue appears to be an issue with the destination
file, not the source table files that he is copying.  Your issue is with
backing up the source table files externally.
